In my mind they aren't lovers until someone writes it in a fic. What can I say, I'm a whore for canon (this is also why all my currently set fics are first time stories).
They declared their feelings for each other sometime in S1 (after Jitters), but they didn't consummate their love* until after Clark either (a) turned 18, or (b) graduated from high school (since high school in the SV-verse starts at 15/16 and ends at 19/20).
*This was due to a variety of factors -- Clark's age, Bo's shotgun, and Lex's inability to believe that he was capable of loving someone, to name a few.
Three minutes before Jonathan Kent showed up to take Clark home after Lex ran him off the bridge. *giggles* Perfect timing...they were already all wet and messy...they could blame it on the 'shock'...Clark luckily had a bottle of apple pie-scented lotion in his pocket...

I like to think that they danced around each other and were just about to declare their love for each other when Lex disappeared ... therefore they hooked up when Lex came back

I went with after Clark became 18, only because, whilst I love young and innocent Clark I do love fics in which they wait until Clark is 18 before actually having sex out of respect for Ma Kent (not so much Pa) or to safeguard Lex's political ambitions. Plus I love Clark whining and begging for sex and Lex being all superior and calmly stating his reasons for abstaining then locking himself in the bathroom for half an hour! LOL
I think I personally like them dating after Lex loses his memory though. Cos a)he's vunerable and b) Clark should have felt REALLY fucking guilty and that more than anything else would make him examine his feelings and the reasons he has for holding back.

I think any of those could have been the moment, but I'm also not sure that any of them were, although I'm of the camp that's persuaded fic-by-fic. In canon, I think they wouldn't be so miserable with each other now if there weren't seven years of UST between them. Really, if they'd just shagged early on, the world would be a happier place for all of us. ;)
